Renal disease tissue binding

The polymyxins are slowly excreted by glomerular filtration the slow elimination rate is due to binding in tissues. Elimination is decreased in patients with renal disease, and drug accumulation can lead to toxicity. Sodium coUstimethate, the parenteral preparation, binds less to tissue and is excreted faster than the free base. [Pg.554]

Altered tissue binding may also affect the apparent volume of distribution of a drug. For example, the distribution volume of digoxin has been reported to be reduced by 30% to 50% from normal values in patients with renal disease. It has been postulated that this reduction in the distribution volume is secondary to a decrease in tissue... [Pg.921]
