The following protocol assumes that the amine-containing oligo has already been synthesized by any of the methods discussed in Section 2.1, this chapter. 

Amide bond linkage resulting in 5 -biotinylated oligonucleotide 

Prepare 10-20 pg of amine-containing oligonucleotide in 200 pi of water. Add to this solution, 20 pi of 1M sodium bicarbonate, pH 9.0.
