The fundamental parameters for bioanalytical validations include accuracy, precision, selectivity, sensitivity, reproducibility, stability of the drug in the matrix under study storage conditions, range, recovery, and response function (see Section 8.2.1). These parameters are also applicable to microbiological and ligand-binding assays. However, these assays possess some unique characteristics that should be considered during method validation, such as selectivity and quantification issues. 

Ion chromatography high-performance liquid chromatography (HPLC) is the method of choice for the quantification of oxalate, glycolate, and glycerate [3,4]. In addition, preservation and storage of the liquid samples may influence the stability of oxalate and glycolate. Use of urinary filter spots is a practical alternative approach for the collection and safe transport of samples to be analyzed for many metabolic disorders [5].

Stability of the analyte during the whole analytical procedure is a prerequisite for reliable quantification. Therefore, full validation of a method must include stability experiments for the various stages of analysis including storage prior to analysis. 

Because carotenoids are labile pigments, their stability on storage in foods and tissues and during their chemical isolation and quantification poses a problem. Carotenoids are especially sensitive to light, heat, oxygen, acids, and, for some compounds, to alkaline environments (Simpson and Chichester, 1981).
