The molecular mechanism of TAP inhibition by US6 is poorly defined mostly because of the lack of isolated viral protein. By use of photoactive peptides it was shown that US6, in contrast to ICP47, does not prevent peptide binding to TAP (Ahn et al. 1997 Hengel et al. 1997). The analysis of chimeras with E3/19K or CD8, as well as of a carboxy-terminal truncation, led to the conclusion that the luminal domain, US6(20-139), is still functional (Ahn et al. 1997). [Pg.94]
