Protein digestion observed

The positive effect of convection of the substrate solution on mass transfer can be observed even better with macromolecular substrates that undergo processes such as protein digestion. For example, Fig. 9 compares reversed-phase chromatograms of cytochrome c digests obtained by cleavage with trypsin immobilized in both packed and molded column reactors, and clearly demonstrates the much higher activity of the monolithic device under otherwise similar circumstances [90].

Similar to the potential utility of patterns generated from mass spectra analyses, patterns of protein expression observed by 2D-PAGE may also be used for diagnostic purposes [6, 46, 47]. 2D-PAGE resolves complex protein mixtures into numerous unique spots, forming a distinctive pattern. The pattern formed by the protein spots from normal and diseased tissue can then be visually compared. Proteins of interest can be excised from the gel, digested using trypsin, and analyzed by MS to reveal their identity. 

When protein reacts with oxidizing lipids, extensive loss of available tryptophan is observed. This loss is due to the destruction of this amino acid and to the general decrease in protein digestibility.46 48... 

In a preliminary study, we compared the reproducibility and peptide distribution across eight salt fractions (0 to 2M ammonium acetate, pH = 3.5) for a protein extract of control U937 cells. Figure 8.3a compares the distribution of peptide clusters for individual salt fractions to that observed for the injection of the same sample on a one-dimensional nanoLC-chip-MS system.
