Protein-A-agarose

Equal volumes of antigen and sera were combined in 0.85% (w/v) NaCl. Each mixture was incubated on ice for 7 hr. Protein A agarose was added followed by an additional 1 hr incubation on ice. Protein A was pelleted by centrifugation and the supernatants assayed for activity.

Next morning, add 50 fi of protein A-agarose beads into each tube. Rock for additional hour at room temperature. 

Wash protein A-agarose beads three times with immunomix and three times with PBSc. For each washing step, add 1 ml of washing solution, mix well, spin 20 sec in microcentrifuge, and aspirate supernatant.

Protein-A-agarose. This protein, derived from cell wall of Staphylococcus aureus binds with very high affinity to the region of the immunoglobulin G. It has been extensively used in antibody purification.
