Physiological responses to prostaglandins encompass a variety of effects Some prostaglandins relax bronchial muscle others contract it Some stimulate uterine con tractions and have been used to induce therapeutic abortions PGEj dilates blood vessels and lowers blood pressure it inhibits the aggregation of platelets and offers promise as a drug to reduce the formation of blood clots... 

Operational model, devised and published by James Black and Paul Leff (Proc. R. Soc. Lond. Biol. 220,141-162, 1983), this model uses experimental observation to describe the production of a physiological response by an agonist in general terms. It defines affinity and the ability of a drug to induce a response as a value of x, which is a term describing the system (receptor density and efficiency of the cell to convert an activated receptor stimulus into a response) and the agonist (efficacy). It has provided a major advance in the description of functional effects of drugs see Chapter 3.6 for further discussion.

The steroids are a remarkable family of molecules. They are essential for health, responsible for the characteristics of our genders, useful medicinal agents, and drugs of abuse. Modest structural variations on the central theme of steroid structure profoundly alter the physiological response to the action of these molecules. Small differences in structure can create big differences in function. 

Although this type of drug has various subclasses based on physiological response, such as tranquilizers, stimulants, depressants, etc., we will subdivide and treat a few of them on the basis of their chemical classes. The leading antidepressant, sertraline (no. 10), was covered in Section 3.10. 

The adrenomimetic drugs, including the naturally occurring catecholamines, initiate their responses by combining with a-, P-, or dopamine adrenoceptors. This interaction triggers a series of biochemical events starting within the effector cell membrane that eventually culminates in the production of a physiological response. 

We have worked in the area of applications for pH sensitive materials, particularly in the area of the controlled release of drugs. Our group s work focuses on physiologically responsive hydrogels, and reviews of this topic have been presented [2, 35].
