Photoaffinity linker

Put the glass slides on a Teflon slide chamber facing down. Fill the space between the chamber and the glass slides with photoaffinity linker solution. Cover the chamber with a glass lid. 

PNP-DTP has been used to photoaffinity-label the thyroid hormone nuclear receptors in intact cells by preparing a derivative of 3,5,3 -triiodo-L-thyronine with the cross linker (Pascual etal., 1982 Casanova etal., 1984). Effective photoreactive conjugation was found to occur after irradiation with UV light at 254 or 310 nm.

Amine-coated glass slides were derivatized with a linker terminated with the known photoaffinity reagent tritiuoromethyldiazirinbenzoyl (Fig. 6), which upon UV irradiation releases a nitrogen molecule and is converted into a highly reactive carbene that reacts indiscriminately with small molecules.
