Notably, only very few directed, nonpolar contacts were found in the comparison of A-butyl-1-deoxynojirimycin (131, Scheme 33 PDB 2V3D) and its A-nonyl analogue (132 PDB 2V3E) in the active site of glucocerebrosidase.269 Neither their numbers nor the difference between the individual contacts for compounds 131 and 132 readily explains the striking difference in biological activity when compared to each other as well as to the parent compound 2.
