MPB-DPPE

Dissolve a sulfhydryl-containing peptide hapten at a concentration of 25 pmol/ml in degassed, nitrogen-purged lOmM HEPES, 0.15M NaCl, pH 7.0. Add the peptide solution to the liposome suspension at a molar ratio necessary to obtain at least a 5 1 excess of thiol groups to the amount of maleimide groups present (as MPB-DPPE).

Prepare a liposome suspension, containing MPB—PE, at a total lipid concentration of 5 mg/ml in 0.05 M sodium phosphate, 0.15 M NaCl, pH 7.2. Activation of DPPE with SMPB is described in Section 2. A suggested lipid composition for vesicle formation is PC cholesterol PG MPB—PE mixed at a molar ratio of 8 10 1 1. The presence of relatively high levels of cholesterol in the liposomal recipe dramatically enhances the conjugation efficiency of the component MPB—PE groups (Martin et al., 1990). Any method of emulsification to create liposomes of the desired size and morphology may be used (Section 1). 

Working table for making a 30 mg of ultrasound-sensitive liposomes correspond to 69 8 8 15 molar percent of Egg PC, DPPE DPPG CH. For conjugation, DPPE is substituted by MPB-PE or PEG2000-PE, otherwise composition remains the same... 

MPB-PE or PEG-PE be used to replace DPPE when antibody conjugation is needed.
