Moxifloxacin dosage

When administered orally, moxifloxacin is absorbed rapidly and almost completely, and has a bioavailabihty of approx. 91% and a plasma half-life of approx. 12 h, which allows once-daily dosing. The plasma protein binding value of 40-42% is low [223]. Moxifloxacin penetrates rapidly into the tissues and body fluids and forms high concentrations in the target tissue these remain higher than the MIC values for the most important pathogens over the entire dosage period of 24 h [199,224,225]. 

Slight to moderate renal dysfunction does not significantly affect the pharmacokinetics of moxifloxacin and its metabolites hence, there is no need for dosage adjustment under such circumstances [226]. Although slight to moderate liver dysfunction tends to reduce the plasma concentrations and renal secretion of moxifloxacin, these do not affect the drug s safety or efficacy [227]. 

Quinolones. Levofloxacin, ciprofloxacin, and moxifloxacin are sometimes used to treat MDR-TB. Moxifloxacin also is being studied as a possible replacement for certain first line agents, but data are not available at this time. Quinolones are useful because most are available in oral and intravenous dosage forms, so they can be used in critically ill patients. 

The increased levels and decreased renal excretion of clinafloxacin caused by probenecid are not considered large enough to warrant dosage adjustment," and most of the changes seen with the other quinolones were of a similar magnitude. However, caution has been advised in the presence of other drugs that may also compete for renal excretion (such as some penicillins or cephalosporins). " Grepafloxacin, moxifloxacin, sparfloxacin, and probably ofloxacin, appear not to interact.
