Monitoring, nucleotide synthesis

Even when the polymer is insoluble, the determination can be made. For example, the extent of reaction in a nucleotide synthesis can be monitored by determining the decrease in nucleotide concentration in the supernatant solution by uv absorption. Alternatively, the polymer-bound nucleotide concentration in the supernatant solution can be determined by uv absorption. Alternatively, the polymer-bound nucleotide concentration in the supernatant solution can be determined by uv absorption. [Pg.42]

Lynen had studied chemistry in Munich under Wieland his skill as a chemist led to the successful synthesis of a number of fatty acyl CoA derivatives which proved to be substrates in the catabolic pathway. Many of these C=0 or C=C compounds had characteristic UV absorption spectra so that enzyme reactions utilizing them could be followed spectrophotometrically. This technique was also used to identify and monitor the flavoprotein and pyridine nucleotide-dependent steps. Independent evidence for the pathway was provided by Barker, Stadtman and their colleagues using Clostridium kluyveri. Once the outline of the degradation had been proposed the individual steps of the reactions were analyzed very rapidly by Lynen, Green, and Ochoa s groups using in the main acetone-dried powders from mitochondria, which, when extracted with dilute salt solutions, contained all the enzymes of the fatty acid oxidation system. [Pg.118]

The formation of pectin and its assembly for final export across the plasmamembrane will be considered here as a production line to indicate the various limiting steps which are controlled to monitor its production. The various channels for the movement and synthesis will be described separately so that the control points can be assessed. The channels which operate for production and movement of the polymers within the cytoplasm and endomembrane system of the cells are - Channel 1. Production and movement of nucleotide sugar donors. Channel 2. Synthesis and compartmentalization of the pectin polymers within the endomembrane system. Channel 3. Movement of vesicles and fusion with the plasmamembrane for assembly and deposition within the wall. [Pg.135]

AMP-activated protein kinase (AMPK) monitors cellular energy, regulates genes involved in ATP synthesis and consumption, and is allosterically activated by nucleotides and synthetic ligands. Mammalian AMPK is a 134-150 kDa heterotrimeric complex composed of a catalytic a subunit that... [Pg.210]

These methods do not find wide, general applications in monitoring of solid-phase synthesis. Some of the polymer-mediated syntheses have been carried out using a soluble polymer support. Nucleotides show characteristic uv absorption, and the attachment of a nucleotide residue to a soluble polymer is easily followed by separating it by ultrafiltration or precipitation. The polymer-bound nucleotide is then redissolved and the concentration of the bound nucleotide determined by uv absorption (Hayatsu and Khorana, 1967). [Pg.42]
