The ratio between pharmacokinetic measures, such as the plasma minimum concentration (Cmin), and the 50% inhibitory concentration (ICj ) provides a measure of systemic antiviral efficacy. Similarly, the ratio of CSF concentrations to IC, may provide a better estimate of CNS efficacy than CSF concentration alone. 

Knowing the Clp and ty2 of a drug one can modify the size and frequency of doses. Doses should be given at repeated intervals such that the rate in equals the rate out . If doses are too high and frequent, then toxic levels may be achieved. If doses are too low and infrequent, then the drug may prove ineffective. Figure 4 shows the ideal situation where plasma concentrations reach a plateau and the same maximum (Cmax) and minimum (Cmii) concentrations are reproduced within the therapeutic window, i.e., Cmin is kept above the minimum effective concentration and Cmax below the minimum toxic concentration.
