Activity in these tricyclic compounds is restricted to compounds having a two- or three-carbon side chain and methyl-substituted or unsubstituted amino groups in the side chain. Some compounds with substituents on the aromatic ring are active. Finally, the two-carbon bridge linking the aromatic rings may be -CH2—CH2— or -CH=CH—. Amitriptyline is recommended for the treatment of mental depression, with improvement in mood seen in two to three weeks after the start of medication. Imipramine is used in similar cases. 

In the second scheme, the specified reaction of amitriptyline (7.1.4) with ethyl chloroformate leads to the substitution of a methyl group on the amino group with an ethoxycar-bonyl group. Hydrolysis of the formed product leads to nortriptyline (7.1.17) [26].

Melitracen is structurally and pharmacologically related to imipramine, with two methyl groups attached to the central ring. It has similar efficacy to amitriptyline, with a somewhat more rapid effect and similar adverse effects (1).
