Method for selecting nutrient medium

PRELIMINARY PROCEDURE METHOD FOR SELECTING NUTRIENT MEDIUM [Pg.94]

The next step is choosing a nutrient medium. Amino acids supplied by high serum concentrations may enhance cell growth significantly in deficient media. As the amount of serum is decreased, the need for a balanced nutrient medium becomes more critical. [Pg.94]

Use the serum lot, serum concentration and cell density chosen in the previous experiment. This should provide the most stringent test of the nutrient balance of the medium. [Pg.94]

Use the type of medium the cells have been growing in as control. Test 3-4 commercially available balanced media, such as F12/DMEM (Ham s nutrient mixture/Dulbecco s modified Eagle s medium), DMEM, IMDM (Iscove s modified Dulbecco s medium), HB-CHO (Hana Biological Chinese hamster ovary medium) or RPMI-1640 (Roswell Park Memorial Institute). [Pg.94]

Use STI if trypsin is used to passage the cells and centrifuge the cells once to remove residual trypsin and STI. [Pg.94]
