Crystal violet (20) can be produced directly from formaldehyde and dimethyl-aniline. The methane base (18), WAfW,W-tetramethyl-4,4 -mcthylcncdianilinc, is formed initially. This is then oxidized to Michler s hydrol (12), which condenses with another molecule of dimethylaniline to give leuco crystal violet (19). The latter is converted to the dye in a second oxidation step. Pararosaniline, methyl violet, and Victoria blue can also be obtained by this reaction sequence. [Pg.64]

Adenosine triphosphate, when oxidized to its dialdehyde with periodate and condensed with Sepharose-adipic (or sebacic) acid hydrazide, has been used to isolate poly(A) polymerase, and also myosin and its fragments from different sources. A poly(A)-linked resin has been prepared by condensing 4,4 -diaminodiphenyl-methane with periodate-oxidized starch, reducing the resulting Schiff bases with borohydride, diazotizing, and then coupling with the polynucleotide. The original method involves dibromination of the cyclobutanone and consequently is not suitable for substrates containing isolated double bonds. In this case an alternative approach is available via a-trimethylenedithiocyclobutanones. Direct condensation of trimethylene dithiotosylate (4, 539-540 5, 71) with the cyclobutanone enolate fails, but can be accomplished indirectly by conversion of the cyclobutanone into an enamide by reaction with i-butoxybis(dimethylamino)-methane. The desired dithiane is then obtained by reaction of the enamide and trimethylenedithiotosylate in ethanol buffered with potassium acetate. The sequence is illustrated for 1-tetralone (1).
