One of the compoimds commonly determined as its Phase 1 metabolite is benzene, whieh is metabolized to phenol in the body (see Chapter 23, Section 23.13 and Figure 23.16). The phenol metabolic product of benzene is measured in urine, usually by gas chromatography. The urine sample is treated with perchlorie aeid to hydrolyze phenol conjugates and the phenol is extracted into diisopropyl ether for ehromatographic analysis. Two other metabolic products of benzene, trans,trans-mueonie aeid and S-phenyl mercapturic acid, are now commonly measured as more-speeifie biomarkers of benzene exposure.

The use of paper and thin-layer chromatography has extended the detection of mercapturic acids to trace amounts in the urines. The problem of interfering substances is largely obviated by the resolving power of the separation procedures. In a few oases, determination of the area of the spot on the chromatogram has provided a semiquantitative estimation of the amount of metabolite excreted (Bray et aL, 1964). 

Early studies on the metabolism of mercapturic acids involved determination of the sulfur distribution in the urine as sulfate, neutral sulfur, and ethereal sulfate. The procedures used were incapable of detecting minor metabolites. Changes in the neutral sulfur were assumed to arise from excretion of mercapturic acids particularly.
