Neri et alF describe the used of the water-soluble nitroxide spin label, TEMPOL, to map the solvent-exposed surface of hen egg-white lysozyme. Pintacuda and Otting have used the Gd chelate, Gd(DTPA-MBA), for the same purpose in studies of ubiquitin. The latter group reports that the correlation of maps generated using TEMPOL and the Gd agent were poor. They recommend the use of the latter, which is more strongly relaxing and less hydrophobic, hence less likely to bind to specific hydrophobic patches of the protein surface. [Pg.576]
