The method used for detection depends on the type of label used. Isotopic counting is employed for radioisotopes, colorimetry for enzyme assays, luminescence and fluorescence measurements can be achieved by means of photomultiplier tubes, while turbidimetry or nephelometry is used for particle enhanced assays. 

Chromogenic reagents include the various kinds of chemicals used as pH indicators (for both aqueous and nonaqueous systems), redox indicators, metal indicators, giving changes usually in color but also in fluorescence or luminescence, and reagents used for visual and/or absorptiometric colorimetry. 

Systems using colorimetry tend to be less sensitive than luminescent systems, but they do have some advantages. By producing a precipitate associated with the DNA-probe hybrid they are ideal to produce a permanent record of the distribution on a membrane. 

Since 1931, the most widespread color spaces for colorimetry of luminescent materials are based on the CIE XYZ color space.
