MS analysis of lipid A from laboratory-adapted PA strains grown in rich medium reveals a dominant ion species (m/z) of mass 1447 (Fig. 11,2a), corresponding to a penta-acylated molecule. This is the major structural form of lipid A isolated from laboratory-adapted PA strains (PAOl, PAK, PAM) grown in rich medium (Bhat et al., 1990 Ernst et al., 1999 Moskowitz et al., 2004). Additional lipid A species observed by MS are dependent on growth conditions (concentration of magnesium or iron, low pH, and reduced oxygen levels), and the strain from which the lipid A was isolated (Goldman et al., 1988). In PA strains, penta-acylated lipid A species are usually most abundant however, specific lipid A structures often differ between CF (m/z 1447, Fig. 11.2a) and non-CF isolates (m/z 1419, Fig. 11.2d). [Pg.245]

Soapstock is recovered via the alkaline refinement process of oilseeds and contains phosphorus lipids, hydrateable and nonsaponifiable compounds, soaps of FFA, vitamins A and E, as well as carotenoid pigments. In their work, condensed corn-soluble basal medium was initially fed with biodiesel glycerol water but when the dissolved oxygen level rose above 10%, the medium was fed with sunflower soapstock. A shift in the monomer compositions from 3-hydroxydecanoate to 3-hydroxyoctanoate was observed when the medium was fed with sunflower soapstock. The final PHA also contained 3-hydroxytetradecanoate and 3-hydroxytetradecenoate monomers with a CDW and PHA content of 17 g/1 and 17%, respectively. [Pg.38]

Although atherosclerosis and rheumatoid arthritis (RA) are distinct disease states, both disorders are chronic inflammatory conditions and may have common mechanisms of disease perpetuation. At sites of inflammation, such as the arterial intima undergoing atherogen-esis or the rheumatoid joint, oxygen radicals, in the presence of transition-metal ions, may initiate the peroxidation of low-density lipoprotein (LDL) to produce oxidatively modified LDL (ox-LDL). Ox-LDL has several pro-inflammatory properties and may contribute to the formation of arterial lesions (Steinberg et /., 1989). Increased levels of lipid peroxidation products have been detected in inflammatory synovial fluid (Rowley et /., 1984 Winyard et al., 1987a Merry et al., 1991 Selley et al., 1992 detailed below), but the potential pro-inflammatory role of ox-LDL in the rheumatoid joint has not been considered. We hypothesize that the oxidation of LDL within the inflamed rheumatoid joint plays a pro-inflammatory role just as ox-LDL has the identical capacity within the arterial intima in atherosclerosis. [Pg.98]
