This spectroscopy has been used as a sensitive and effective analytical technique for the characterization of specific ligand-oligonucleotide duplex non-covalent complexes [138], due to its specificity, sensitivity and quickness as well as its advantage in determining binding stoichiometry. [Pg.172]

Conceptually the most simple syntheses of complex molecules involve the joining of structural units in which all functional groups and all asymmetric centres are preformed. This technique can usually only be applied to compounds in which these units are connected by —C—X— bonds rather than C—C. Critical to the success of this procedure was the development of a suitable temporary ligand that is sufficiently inert to conditions of oligonucleotide deprotection (typically aqueous ammonia, 6h at 50° C) and can subsequently be replaced by chloride. The nucleobase derivative 1-cyclohexamethyl-thymine 41 was found to satisfy these criteria and can be converted to the trans-Pt-Cl species by treatment with dilute HC1 (pH 2.3, 40°C, 48 h).
