The physiological effects of the sulphidopeptide leu-kotrienes, including LTC4, are potent contraction of bronchial smooth muscle, contraction of arterial, arteriolar and intestinal smooth muscle, enhanced permeability of post-capillary venules and enhanced bronchial mucus secretion. Because of their potent effects on the airways, leukotrienes have long been regarded as important molecules in the pathogenesis of asthma. Because of this, and the multicellular provenance of leukotrienes many comprehensive reviews have been written on these molecules to which the reader is referred (Ford et al., 1994 Chanarin and Johnston, 1994 Valone et al., 1994). [Pg.61]

Pretreatment with LYl 71883 prevented the endotoxin-induced neutropenia (Figure 5.13), haemoconcentration (Figure 5.14) and acute systemic hypotension observed in rats . The increased haemoconcentration of endotoxic shock, an indirect measure of vascular permeability, suggests enhancement of vascular permeability by endogenous LTD4/E4. It is not clear whether the prevention of neutropenia by LYl 71883 in endotoxaemia is a direct effect of inhibition of the action of sulphidopeptide leukotrienes. For example, these leukotrienes have been shown to stimulate neutrophil adherence to cultured endothelial cells by a platelet activating factor-dependent mechanism. ... [Pg.113]
