Lansoprazole Foods

Agents in this class are omeprazole, lansoprazole, pantoprazole and rabeprazole. Esomeprazole is the S-enantiomer of omeprazole. After ingestion of gastric acid resistant formulations they are rapidly and more or less completely absorbed. Bioavailability may be reduced if administered with food or antacids. Elimination is via metabolism in the liver and the renal excretion of inactive metabolites. The elimination half-live is very variable, however, as explained above, not related to the duration of action. [Pg.379]

Food modestly reduces the bioavailability of lansoprazole and esomeprazole, but not omeprazole, pantoprazole, or rabeprazole. [Pg.970]

There are some differences in the effect of food on oral bioavailability. Generally, a foregoing breakfast causes some delay in appearance of the drugs in plasma. Bioavailability (AUC) of omeprazole remains unchanged [13], whereas that of lansoprazole is markedly reduced [16,17]. No alteration of bioavailability was found with rabeprazole [18] and in the majority of cases with pantoprazole. However, 5 of 24 volunteers displayed no measurable plasma concentrations for a period of 8 h after oral intake [8]. Tentatively, this finding may be ascribed to pharmaceutical rather than drug-specific properties. Although an effect of food was not obtained in a further study with lansoprazole, application before breakfast is emphasized with this PPI [19]. [Pg.145]
