Km—Reaction Coordinate Diagrams

Uniform binding of substrate and transition state imparts no extra catalysis 

We find experimentally that many enzymes have Km values nearly equal to or slightly greater than the natural concentration of their substrates. Often binding is just strong enough to have a high fraction of the E S complex present at equilibrium.
