The student who is unacquainted with the study of enzymes should first read the following brief theoretical introduction. 

The use of mass spectrometry for the analysis of peptides, proteins, and enzymes has been summarized. This chapter should be read in conjunction with others, including Chapter 45, An Introduction to Biotechnology, and Chapters 1 through 5, which describe specific ionization techniques in detail. 

Plasmid Vectors for Facile Introduction of Passenger DNA and Selection of Recombinants. The map of a commonly used plasmid vector, pUC19 (7), is shown in Figure 2. As described earlier, translation of the EPSPS mRNA of plants results in the formation of a protein which has an AJ-terminal extension. The AJ-terminal extension, referred to as the chloroplast transit peptide, is necessary and sufficient for the import of the preprotein by the chloroplast. Once imported by the chloroplast, the transit peptide is cleaved releasing the mature enzyme. As expected, introduction of the EPSPS transit peptide to other protein sequences results in the importation of the fusion protein by the chloroplast. 

Earlier formulations contained mainly chlorine bleach, metasiUcates, triphosphate, and nonionic surfactants. Modem manufacturers have switched to more compHcated formulations with disiUcates, phosphates or citrate, phosphonates, polycarboxylates, nonionic surfactants, oxygen bleach, bleach activator, and enzymes. The replacement of metasiUcates by disilicates lowers pH from approximately 12 to 10.5, at 1 g ADD/L water. The combined effect of decreased pH, the absence of hypochlorite, and the trend toward lower wash temperatures has paved the way for the introduction of enzymes into ADDs. Most ADD brands in Europe are part of the new generation of ADD products with enzymes. The new formulations are described in the patent hterature (55—57). 

The design of potent inactivators requires the determination of the functional groups necessary for optimal binding of the inhibitor to the enzyme. Next, the regions of steric tolerance are identified that will be useful for introduction of the reactive group. The reactivity as well as the size of the reactive...
