Insulin-related material

Extraction of Insulin-Related Material and Other Peptide Hormones from Tetrahymena pyriformis... [Pg.177]

Following the extraction of the cells in acid-ethanol, the material was applied to Sep-Pak C- q cartridges and eluted by a discontinuous acid ethanol gradient. The insulin-related material was eluted in the 60 and 70 ethanol eluates (Figure 3). [Pg.181]

Figure 3. Sep-Pak (C g) purification of insulin-related material. I-insulin and purified porcine insulin were added to separate batches of Tetrahymena pyriformis homogenates and applied to Sep-Pak cartridges. Following elution from the Sep-Pak each gradient fraction was tested for recovery of the exogenously added insulin. The results for each fraction are expressed as the percent of the total material recovered. Key , endogenous insulin Tetrahymena , 125i-insulin added to extract 0, porcine insulin added to extract. Insulin-, somatostatin-, ACTH-, and relaxin-related materials were detected in the partially purified extracts using specific double antibody radioimmunoassays (12,8,16,7). [Pg.179]

AMP is related to certain mental diseases and may be involved in the action of tranquilizers and antidepressant drugs (60). Whether the ability of diuretic agents such as ethacrynic acid and chlorthalidone to inhibit the enzyme in kidney (68) is related to their diuretic action is also not known. It has been suggested that inhibition of diesterase by diazoxide (59) may explain the hyperglycemic activity of this agent. Several materials are known to activate the enzyme. Imidazole produces strong activation of the enzyme from mammalian tissues (36, 38, 42) but not from E. coli (41). It has been reported (61) that insulin activates the beef heart enzyme in vitro, but it is not known if this has relevance... [Pg.369]
