Imidazole 1.2- diphenyl-5-ethyl

Ethyl acetate and ethyl propionate in ethanolic sodium ethoxide with 5-aminoimidazole-4-carboxamide and its riboside have provided a route to 2-methyl- and 2-ethyl-hypoxanthine arid corresponding 2-alkylinosines, respectively (67JOC3258,68JA2661). 2-Methylhypoxan-thine has also been prepared from the same imidazole and ethyl orthoacetate via an intermediate ethoxymethylene derivative which cyclized to the purine when heated (60JA3144). The same method has been applied to the synthesis of 2,8-dimethyl- and XS-diphenyl-hypoxanthine from appropriate imidazole precursors.

Photolysis of 2,3-diphenyl-A -azirine (442) generates benzonitrile ylide (443). Irradiation in the presence of ethyl cyanoformate resulted in a mixture of the oxazoline (444) and the imidazole (445) by 1,3-dlpolar cycloaddition to the carbonyl and nitrile group, respectively (72HCA919).

Condensation of aryl halides with various active methylene compounds is readily promoted by catalytic action of palladium to give the corresponding arene derivatives containing a functionalized ethyl group [7]. Yamanaka et al. extended this chemistry to haloazoles including oxazoles, thiazoles and imidazoles [8]. Thus, in the presence of Pd(Ph3P)4,2-chlorooxazole was refluxed with phenylsulfonylacetonitrile and NaH to form 4,5-diphenyl-a-phenylsulfonyl-2-oxazoloacetonitrile, which existed predominantly as its enamine tautomer. In a similar fashion, 4-bromooxazole and 5-bromooxazole also were condensed with phenylsulfonylacetonitrile under the same conditions.
