IkBa degradation

In the NF-kB pathway with celastrol 76, the inhibition of the iKBa degradation is due to the upstream blockage of the kinase activity and not by the direct inhibition of proteasome activity. On the contrary, direct inhibition of proteasome activity was observed with celastrol 76 and pristimerin 2 in prostate cancer cells.90-92 Both triterpene QMs directly inhibited the activity of the 20S subunits of proteasome at 2.5 iM and induced the accumulation of ubiquitinated proteins over time in cells,... [Pg.284]

When TRAF2 and RlPl bind to the complex, two sequential pathways are activated the NFkB pathway and the activated caspase-8 pathway. In the first step of the NFkB pathway, TNF activates the IxBa pathway in a process that depends on the degradation of the inhibitor IkB by the proteasome. The Ikk complex (IkB kinase) mediates the phosphorylation of the inhibitor IkB. The Ikk complex is formed by two related IkB kinases, iKBa and kBfl, and NFkB essential modulator (NEMO), a regulatory protein also known as kBy. The roles of TRAF2 and RIP in the Ikk complex are recmitment and stabilization, respeetively (Devin et al., 2003). [Pg.24]

The CG oleandrin is found to block tumor necrosis factor (TNF)-induced activation of NF-kB in a concentration- and time-dependent manner [25]. Oleandrin blocked NF-kB activation induced by phorbol ester and lipopolysaccha-ride. The inhibitory effect of oleandrin on NF-kB is mediated via suppressing phosphorylation and degradation of I kappa B alpha (IkBa), an inhibitor of NF-kB however, these effects of oleandrin were not cell-type specific, because it blocked TNF-induced NF-kB activation in a variety of cells. [Pg.3751]
