Cellular ceramide is generated either by hydrolysis of sphingomyelin or by de novo synthesis. Sphingomyelin hydrolysis is catalyzed by at least three different sphingomyelinases that are discriminated by their optimal pH acid, neutral, and alkaline sphingomyelinases.
