Sheldon and co-workers have circumvented this problem to some extent by three approaches 46 the use of sulfuric acid to reduce the pH, by addition of ammonium fluoride and by addition of ammonia. As another example, Bright and coworkers investigated the oxidation of cholesterol by cholesterol oxidase (ChOx) within the water core of reverse micelles (that had been formed in SCCO2 by using PFPE surfactant) in the presence of catalase, which was added in order to destroy hydrogen peroxide created within the course of the oxidation reaction (Scheme 12.7) [80]. They presented a comparative evaluation of microemulsion stabilized by AOT in isooctane [81] and microemulsion composed of PFPE in SCCO2 as reaction media for the oxidation reaction. 

Given the sensitivity of Met to oxidation compared with that of other amino acid residues, it is important to be able to routinely identify Met oxidation dming manufacture and stability studies of active substance and vialed protein, and to understand its effect on bioactivity. In forced degradation studies, the protein bio-pharmaceutical may be exposed to low levels of oxidants, such as hydrogen peroxide, peracetic acid, or bleach, to evaluate the sensitivity of the protein in question.
