Hyaluronic acid periodate oxidation

Hyaluronate lyase purified from Streptomyces hyalurolyticus has been used to provide a specific assay for hyaluronic acid, its action resulting in the generation of 8-D-gluco-4-ene-pyranosyluronic acid groups. The assay is based on the long established periodate oxidation of such groups to inter alia j3-formylpyruvic acid which specifically gives a chromophore with thiobarbituric acid.

Teichan was shown to be very labile in presence of hot alkali, which would indicate some difference in its chemical structine from those of hyaluronic acid and chondroitin. The results of the periodate oxidation of teichan were similar to those of hyaluronic acid, whereas infrared spectra studies confirmed the presence of a-glycosidic linkages, already suggested by the positive optical rotation.
