After iv administration to nude mice, the tissue distribution of PEG-rMETase was found to be in the following decreasing order blood, kidney, hver, spleen, heart, lung, tumor, intestine and muscle. Significant PEG-rMETase levels accumulated in the tumor. One hour after iv injection of 60 units of PEG-rMETase, levels were approximately 0.026 units/mg protein in human colon tumor HCT 15 growing subcutaneously in nude mice compared to 0.017 units/mg protein for rMETase. These results demonstrated that PEG-rMETase could accumulate in the tumor at least as well as rMETase. ...
