It should be noted that derivatization with BSTFA requires a completely water-free atmosphere. In the presence of small amounts of water (atmospheric moisture levels), derivatization can be disturbed, and loss of substance can occur. To derivatize the sample, the vial is put on the hot injector for around 45 min. The sample should then be analyzed immediately, as the peak area decreases with increasing time.
