Peripheral tissues such as muscle, adipose tissue, and the lactating breast contain both hormone-sensitive lipase and lipoprotein lipase. This second lipase, mentioned in the lipoprotein section of Chapter 6, requires apolipoprotein C-II as a cofactor. The lipase is secreted by cells and migrates through the interstitial fluid to the capillary, where it becomes bound to the membrane with its active site exposed to the bloodstream. This binding to the luminal wall of the capillary occurs by attachment to a polymer called glycosaminoglycan. [Pg.215]

The first heterogenous RI Awas reported for human growth hormone (HGH) by Catt et al. in 1967 [15] (Fig. 6). Anti-HGH antibodies were immobilized on polystyrene. Plasma containing standard or unknown HGH concentrations together with a probe of radiolabelled HGH was added to the immobilized antibodies, and the hot and cold HGH competed for the available binding sites. Finally, the radioactivity present on the polymer was measured and correlated with the cold HGH concentration the concentration of cold HGH in unknown samples was determined by comparing the bound radioactivity with a standard curve determined for standards of known cold HGH concentrations, similar to the curve shown in Fig. 4b. [Pg.646]
