It has been shown that JHDMIA demethylates only mono- and dimethyl lysine of histone H3 (H3K36). This was rather unexpected given that the catalytic mechanism of a Jmj C domain does not require a protonated nitrogen, unlike LS D1, and therefore could also have the ability to remove the methyl in all three states. So far, two crystal structures have been deposited in the protein database but without any publication underlining the work. The crystal structures describe two forms of the human JHDMIA lysine demethylase, apo and in a complex with a-ketoglutarate (PDB codes 2yul and 2yu2). [Pg.43]

Notably, the model generated contains one factor, E2F that is known to be involved in cell cycle regulation of genes [67]. The model appears to be very selective and only one match per 12 million base pairs of the mammalian sections of the EMBL database was found. Almost all matches (total of 59) are known Histone HI genes, except in the human section where the majority of matches (25) are within anonymous sequences. However, given the extraordinary specificity throughout the mammalian sections, it seems safe to assume that most of the unknown matches within the human database section actually identified new so far unannotated genes, that are subject to... [Pg.152]

Tervo et al. [155] described a successful virtual screening experiment for the class III histone deacylase sirtuin type 2 (SIRT2). Using two databases of commercially available compounds plus a receptor-based pharmacophore query, 4 out of 11 tested molecules showed weak to moderate inhibitory activity. The two most active inhibitors vdth IC50 values of 51 and 91 pM had new scaffolds (Figure 12.14b). [Pg.345]
