Heck reaction bicyclic systems

Grigg s group synthesized a unique bicyclic P-lactam 108 via an intramolecular Heck reaction from 107 [87, 88]. The 7-membered ring was formed via an unusual insertion at C(3) of furan, an aromatic rc-system. 

Several routes to the pyrrolo[l,2-f][l,2,3]triazole skeleton have been described. Intramolecular dipolar cycloaddition of azido-alkenes or alkynes seems to be the most convenient process, although the cyclization efficiency seems to be highly substrate dependent (Scheme 16) <2002JA2134, 2003T1477, 2005SL2187, 2005TL8639>. The formation of this bicyclic system by an intramolecular Heck reaction is an attractive alternative. We continued our work with the reductive Heck and Domino-Heck reactions [1, 7J of new bicyclic compound (3) by treating it with different aryl- and hetaryl-iodides, as a result a series of new epibatidine analogues were synthesized, continuously separated and purificated by column chromatography on silica gel. Treatment of 3 with iodobenzene, 2-iodothiophene, 1-iodonapthalene and 2-chloro-5-iodopy-ridine under reductive Heck conditions gave new compounds 4a-d and 5b, 5d as exo-regioisomers after chromatographic separations. The reactions with iodobenzene and 1-iodonapthalene gave only 5-exo- products. The use of trimethylsily-lacetylene under Domino-Heck conditions provided alkynic bicyclic systems 6e andf. 

A palladium-cataly/ed C-C coupling reaction—the Heck reaction — is used in the construction of bicyclic system 13. Cyclization leads to a q3-alIyl-Pd complex, which undergoes nucleophilic attack by malonic ester anion 12. This in turn leads to formation of the C4 side chain The mechanism of this reaction therefore differs from that of a normal Heck reaction.

An interesting possibility for the construction of bicyclic systems containing one six-membered ring arises when an intramolecular Heck reaction or palladium-catalyzed enyne cycloisomerization [311] to give a vicinal exodimethylenecy-cloalkane is immediately followed by a Diels-Alder reaction (Scheme 8.15 and Scheme 8.19). This sequence is normally conducted in two steps, but may also... 

In another example, Shibasaki and co-workers applied an asymmetric Heck reaction/anion-capture cascade process to the total synthesis of marine sesquiterpenes capnellenols 14 and capnellene 17 (Schemes 13.5 and 13.6). The cascade involved the use of prochiral cyclo-pentadienyl systems for the generation of bicyclic 71-allyl-palladium species, which were then trapped by a suitable nucleophile. Initial studies in the total synthesis of capnellenols described the trapping of the 71-allylpalladium...
