GPCRs transfected

Another research group has applied gold particle tracking to measure movements of the p-opioid GPCR on the surface of GPCR-transfected fibroblasts [30], They describe the pattern observed in Fig. 2-11 as a walking defined diffusion mode . More than 90% of the observed particles displayed this pattern, which consists of rapid... [Pg.30]

FIGURE 2-11 Video-enhanced Differential Interference Contrast (DIC) images of gold-labeled p opioid GPCR on the surface of a GPCR-transfected fibroblast. The white trace is the trajectory of one particle over 2 minutes at 25frames/s. The black trace is the mean square displacement of the particle as a function of time. Reproduced from Figure 1 of [30], with permission. [Pg.31]

In nonactivated cells, once validated, GFP-GPCRs inform on localization of receptors. Indeed, this can be used as an efficient means of identifying effectively transfected cells to clone them. However, there is a danger of prejudice concerning the desirable characteristics of cells to be cloned. GFP-GPCRs transfected into native tissues or used transgenically may hold out a more fruitful use in establishing the cellular localization of receptors. [Pg.153]

Taken together, the enhanced proliferation of KSHV GPCR transfectants and their tumorigenic activity in nude mice supports a role for this receptor in tumorigenesis. [Pg.232]

Hamdan et al. (2002) describe another biogenic amine-responsive GPCR in the tegument of S. mansoni, which specifically binds to and is activated by histamine. This cloned receptor shares approximately 30% amino acid sequence homology with other major amine GPCRs, and, when transfected into mammalian cells exhibited the following characteristics ... [Pg.222]
