Trace amine-associated receptors (TAARs) are related to G protein-coupled aminergic neurotransmitter receptors such as dopamine and serotonine receptors and recognize derivatives of the classical monoamines such as B-phenylethylamine, octopamine, tryptamine, and tyramine. Initially, TAARs had been considered neurotransmitter receptors (Borowsky et al. 2001), but recently an expression in olfactory sensory neurons was shown for several mammalian taar genes, with expression characteristics very similar to odorant receptors (Liberies and Buck 2006). Thus the taar genes were recognized as a fourth GPCR family of olfactory receptors (Buck 2000).

A simple substructure search of the PDB ligand SMILES database yielded all examples of phenols. Three such examples are illustrated in Figure 10.6. Each of these was clearly involved in a different type of environment, information that was captured in the descriptors automatically output by the search. We collected a dataset of human aminergic GPCR inhibitors from the GVK database (GVK DB) [11]. The inhibitory activity against human aminergic GPCR was expressed as the logarithm of the reciprocal IC value (pIC ), where IC represents the micromolar concentration at which 50% inhibition is achieved.

The ECFP 6 ligand descriptors used to construct the full aminergic GPCR inhibitory activity data were also used in the LPLS analysis. After filtering by 0.05 variance cutoff, 149 fingerprints were eligible from a generated total of 36,252.
