Significant purification of certain components by HPLC has been reported by McCluer et al (14). Most of the more successful separations have involved derivatized glycolipids, allowing detection in the UV region. 

Newer ionization methods such as FAB-MS and SIMS have been developed in which derivatization is unnecessary. With the aid of FAB-MS, it has been possible for the first time to obtain data on such structural parameters as molecular mass, homogene. /, sequences, composition branching and linkage. This method has been successfully applied to glycolipid structure analysis (30,31). 

It should be borne in mind that the majority of glycolipid species with or without derivatization were characterized by many other ionization techniques prior to the utilization of ESI-MS. Those kinds of information are well documented in the books written by Murphy [15, 16], and Christie and Han [17]. The interested readers could consult these books for any further information. 

The monosaccharides in glycolipids can also be determined by gas chromatography. The lipids are hydrolyzed with trifluoroacetic acid and then derivatized to an acetylated glyconic acid nitrile. By using this sugar derivative, the chromatogram is simplified because of the absence of sugar anomers (cf. 4.2.4.6). 

Three j8-D-2-acetamido-2-deoxyhexosidases have been isolated from culture filtrates of a Streptococcus species by affinity chromatography on agarose derivatized with oxamic acid or l-thio- -o-galactopyranose. The enzymes (pH optimum 5.0—5.5) hydrolysed synthetic substrates and glycopeptides, but not glycolipids.
