Glucuronide synthesis is the rate-determining step in hepatic bilirubin metabolism. Drugs such as phenobarbital, for example, can induce both conjugate formation and the transport process.

The source of sulphate for conjugation is limited, while the enzymes responsible for sulphate activation and the transfer of sulphate to the acceptor molecule are found in the soluble fraction of the liver (Robbins Lipmann, 1957). The relatively low capacity of the pig to synthesize ethereal sulphates is compensated for by increased glucuronide conjugation. Sulphate conjugation as well as glucuronide synthesis appear to be limited in fish (Mandel, 1971).
