During the experiment it is extremely important to keep an exact time protocol, since the action of calcium is very rapid, and minor variations may change the amount of secreted insulin. When employing 96-well plates, we generally work in rows of 6 wells, using the same conditions for 3 wells. For each row we allocate 50 seconds to change the buffers, which allows us to manipulate 8 rows (48 wells in total or 16 different conditions in triplicate). In our experience many reactions are critically dependent upon the proper choice of solvent and temperature and DMF is not generally the best solvent though it is the commonest. 70-80 C is a good general working temperature. 

In subproject BI, the PRIME system (PRocess-Integrated Modeling Environments has been developed for the experience-based process support at technical workplaces. The usage of PRIME for a certain application domain requires the definition of executable process models relevant to the domain. Generalized work processes from chemical engineering design, represProcess Models, were used as a basis for the creation of such executable models. This transition is described in Sect. 6.2.
