Freeze-Dried Bovine Gametes with Organic Additives

3 FREEZE-DRIED BOVINE GAMETES WITH ORGANIC ADDITIVES [Pg.820]

Native bovine gametes were centrifuged (3000 rpm) for 10 min, residual cells were four times washed by 2.9% sodium citrate with control of the protein content in the supernatant (spectropho-tometrically at i=280 nm). Cells (with residual amount of sodium citrate) freeze-dried (the bulk density of 0.12 g/cm ) included about 2 wt% of residual water. The same cell sample (equilibrated after each addition for 10 min by shaking) was studied with subsequent addition of water, CDCI3, and DMSO (which can be solved in chloroform). Extracellular chloroform was evaporated from the samples for 2 weeks and then an additional portion of DMSO (up to 5 g/g) was added and the system was equilibrated for 2 h (Turov et al. 2008a). [Pg.821]
