The signal/noise ratio in such measurements depends on the mean number of photons that are detected before a transition, and so decreases as the transitions occur more frequently. Raising the light intensity can increase the signal, but tends to cause more rapid photodestruction of the fluorophores, making it necessary to replace the sample at frequent intervals (Sect. 5.8). [Pg.342]
