We conclude that visible pump - midinfared probe spectroscopy on the femto- to nanosecond-time scale yields very valuable information on the structural dynamics of proteins, which is difficult to obtain otherwise. We have shown that the sensitivity of detection in our setup is such that changes of single amino acids can be resolved, and that it is possible to work with proteins having a very long photocyle, such as PYP, which has a photocyle time of about 1 second.
