Danazol is a sex hormone that suppresses the pituitary-ovarian axis by inhibiting output of pituitary gonadotropins has weak, dose-related androgenic activity with no estrogenic or progestational activity. In addition to myelosuppression, estramustine also possesses estrogenic side effects (gynecomastia, impotence, and elevated risk of thrombosis, and fluid retention) and is associated with hypercalcemia, acute attacks of porphyria, impaired glucose tolerance, and hypersensitivity reactions including angioedema. [Pg.248]

Frequent adverse effects include headache (-15% of patients), GI symptoms (e.g., nausea, vomiting, diarrhea, heartburn, flatulence), and rash. More serious reactions include hepatotoxicity, serum sickness reaction, angioedema, and hematologic effects (e.g., leukopenia, neutropenia, punctate basophilia, and monocytosis).
