The characterization of progestins usually starts with the determination of the binding affinity to PR by a standard displacement experiment. Traditionally these binding studies were performed with cytosol preparations of uteri of estrogen-primed rabbits. These preparations are incubated with the test compound in different concentrations and a radiolabeled reference, mostly tritiated progesterone or R5020 (promegestone 2) (Scheme 6.2).
