Enzyme-catalyzed phosphoryl transfer phosphomonoesters

Table 2 lists phosphatases for which the stereochemical outcome has been determined by using chiral phosphomonoester substrates. In such experiments the phosphoryl group is made chiral using lfiO, 170, and lsO, or, a phosphorothioate substrate is used that has a sulfur atom and two isotopes of oxygen in the nonbridging positions. Enzymes which catalyze the reaction type (a) include phosphodiesterases, phospholipases (C and D), nucleotidyl transferases, nucleases, and pyrophos-phokinases. The type (b) reaction involves mainly phosphokinases and phos-phomutases. The hydrolysis of phosphomonoesters (reaction type c) is catalyzed by phosphatases, nucleotidases, ATPases, and so on. Most phosphatases also catalyze the phosphoryl transfer reaction, type (b), if an alcohol is used as an acceptor. [Pg.288]
