Retrosynthetic disconnections of disorazole Al and Cl envisioned the masked fragment 86, which could be synthesized via the Sonogashira couphng C(10)—C(ll) of vinyliodide 84 and enyne 85 in the presence of PdCl2(PPh3)2 [12]. 

Little is knovm about the mechanistic details of isocyanide-allcyne couphngs. However, a useful stoichiometric cychzation of enynes and diynes with isocyanides was developed hy Tamao and Ito,l ] and Buchwald later developed a catal hic variant (Scheme 34).l l The resulting cyclopentenimines may be hydrolyzed to cyclopentenones. A variety of carhocy-clic and heterocychc templates are tolerated in the sequence.
