ELISA microchip

An example of the immobilization on polystyrene beads is the bead-bed micro-ELISA system of Kitamori et al. [403]. The beads were coated with a capture antibody and loaded into the reaction channel of the ELISA microchip (Scheme 4.92a). A further promising development is the implementation of CEIA on microchip devices. In this case the excellent mass detection limit makes it favorable, in contrast to classic ELISA. The use of whole cells and viruses for CEIA applications is also an emerging field.

As an example of microchip-based electrochemical immunoassays, we describe here the protocol established for the analysis of interleukin IB by enzyme linked immunosorbent assay (ELISA) with amperometric detection at the sub-pM level in DiagnoSwiss microfluidic chip called Immuchip . 

Henares et al. [210] developed a multiple enzyme linked immunosorbent assay (ELISA) chip by using capillary-assembled microchip (CAs-CHIP), which involved simple embedding of a 2 to 3 mm length of square capillaries possessing valving and immunoreaction functions into the microchannels fabricated on a PDMS substrate. The authors immobilized different anti-IgGs... 

Instead of the optical labels, enzymes are also used as a labeling material [7]. ELISA is one of the most popular techniques in the conventional immunoassay and suitable in the microchip format. By using a fluorogenic reaction, the fluorescent signal can be taken at the downstream microchannel without interruption by microbeads. In the case of chromogenic reaction, a TLM is useful [8].
