Electrochemical Metal Nanoparticle-Labeled Aptasensors

Another example of electrochemical aptasensor based on Au-NPs as labels for the detection of thrombin is reported by Zheng et al. [28]. The assay was based on a sandwich format, in which the aptamerl (15-mer DNA aptamer with an amino group at its 5 end) was immobilized onto carboxyl functionalized magnetic beads. Such aptamer-coated magnetic beads were used for capturing and separation. Thrombin and Au-NP-labeled aptamerll were then added to [Pg.40]

A signal amplification was obtained by forming a network of thio-cyanuric acid/Au-NPs. The electrochemical oxidation of Au-NPs was performed at -1-1.25 V for 120 s on a glassy carbon electrode. Immediately after the electrochemical oxidation, differential pulse voltammetry was performed resulting in an anal3ftical signal due to the reduction of AuCl , which relates to the amount of the Au-NPs for the sandwich format. [Pg.41]

A detection limit of 8 aM was achieved. To demonstrate the feasibility of this approach, the aptasensor was applied to the detection of thrombin in some plasma samples. [Pg.41]

Indirect amplified electrochemical analysis of aptamer-protein complexes has employed nanoparticles as labels for the development of electrochemical aptasensors. The use of metal nanoparticles as tracers for the analysis of nucleic acid... [Pg.73]
